Rapid and sensitive PCR detection of Vibrio penaeicida, the putative etiological agent of syndrome 93 in New Caledonia ArchiMer
Saulnier, Denis; Avarre, Jean-christophe; Le Moullac, Gilles; Ansquer, Dominique; Levy, Peva; Vonau, Vincent.
Experimental infections of Penaeus (Litopenaeus) stylirostris were performed with a Vibrio penaeicida strain (AM101) isolated in New Caledonia from Syndrome 93 diseased shrimp. Cumulative mortalities resulting from intramuscular injection or immersion of shrimp in bacterial suspensions demonstrated high virulence for this bacterial strain and suggested that V. penaeicida could be the etiological agent of Syndrome 93. The median lethal dose (LD50) for AM101 was 1.3 x 104 CFU (colony forming units) ml-1 by immersion and less than 5 CFU shrimp-1 by intramuscular challenge, with mortality outbreaks at 48 and 22 h after challenge, respectively. A polymerase chain reaction (PCR) detection assay using a primer set designed from the 16S ribosomal RNA gene of V....

Proteinaceous exotoxins of shrimp-pathogenic isolates of Vibrio penaeicida and Vibrio nigripulchritudo ArchiMer
Aguirre-guzman, Gabriel; Labreuche, Yannick; Ansquer, Dominique; Espiau, Benoit; Levy, Peva; Ascencio, Felipe; Saulnier, Denis.
The pathogenicity of two V. penaeicida strains, AM101 and KH-1, with different geographic origin, and V. nigripulchritudo strain AM102, were investigated in juvenile blue shrimp species Litopenaeus stylirostris. Alive bacteria and protein fractions (PFs) obtained from cell-free supernatants (CFS) were used in experimental challenges. Strains AM102, AM101, and KH-1 produced respectively 60, 54 and 12% mortality at 96h after infection using 104 cfu mL-1 of bacterial suspension. Exocellular toxin-like factors were evidenced in CFS from the New Caledonian strains (AM102 and AM101) but not the Japanese strain (KH-1). At 48h post injection of each CFS, mortality rates were respectively 96,98 and 5% when these strains were cultivated at 20°C, whereas only O,16...

Vonau, Vincent; Rouxel, Catherine; Saulnier, Denis; Cochennec-laureau, Nathalie; Nedelec, Georges; Goyard, Emmanuel.
The tropical seabass Lates calcarifer was introduced in Tahiti in 1984 and then was domesticated without other introduction. Three successive generations have been obtained in captivity. Tissue samples of the 38 animaIs which represent the total Tahitian broodstock were preserved in alcohol to be genotyped with four microsatellite markers (Yue et al., 2001). Three of four described markers were successfully transferred to the laboratory of genetics of Tahiti (marker LcaM01, LcaM02, LcaM03), as the revelation technology available locally did not allow the use of LcaM04 in routine. The results show that the genetic diversity of the tahitian broodstock is not equivalent of the one of the species, which could be explained by the small size of the founder...

Cochennec-laureau, Nathalie; Levy, Peva; Saulnier, Denis; Fougerouse, Angelique.

Vibrio crassostreae sp nov., isolated from the haemolymph of oysters (Crassostrea gigas) ArchiMer
Faury, Nicole; Saulnier, Denis; Thompson, F; Gay, Melanie; Swings, J; Leroux, Frederique.
Polyphasic analysis of five new Vibrio isolates originating from the haernolymph of diseased cultured oysters is described. The new isolates were closely related to Vibrio splendidus, having 98 % 16S rRNA gene sequence similarity. gyrB phylogenetic analysis, fluorescent amplified-fragment length polymorphism (FAFLP) fingerprinting and DNA-DNA hybridization experiments clearly showed that the new isolates form a tight genomic group that is different from the currently known Vibrio species. It is proposed to accommodate these isolates in a novel species, Vibrio crassostreae sp. nov. (type strain LGP 7(T) =LMG 22240(T)=Clp 108327(T)). Phenotypic and chemotaxonomic features that differentiate V. crassostreae from other known Vibrio species include arginine...

Pathogenicity of Vibrio penaeicida or white shrimp Litopenaeus vannamei: a cysteine protease-like exotoxin as a virulence factor ArchiMer
Aguirre Guzman, G; Ascencio, F; Saulnier, Denis.
The pathogenicity of Vibrio penaeicida Strains KH-1 and AM101, their culture-free supernatant (CFS), and their protein fraction obtained by 40% of ammonium sulfate precipitation (PFs40) were assessed in experimental challenges against juvenile Litopenaeus vannamei. Live Vibrio cells, CFS, and PFs40 from the AM101 strain produced a significantly higher mortality (p < 0.05) compared to the KH-1 strain. Toxicity and median lethal doses (LD50) of Fast Protein Liquid Chromatography (FPLC) products were evaluated on L. vannamei. The first FPLC fraction sample (A) from PFs40 of the AM101 strain displayed LD50 values of 1.68 and 5.61 mu g protein ind.(-1), respectively. The second FPLC process from Fraction A showed a peak (A1) also with toxic effects to...

Molecular epidemiology of Vibrio nigripulchritudo, a pathogen of cultured penaeid shrimp (Litopenaeus stylirostris) in New Caledonia ArchiMer
Goarant, Cyrille; Reynaud, Yann; Ansquer, Dominique; De Decker, Sophie; Saulnier, Denis; Leroux, Frederique.
A collection of 57 isolates of Vibrio nigripulchritudo from either diseased or healthy shrimp and from shrimp farms environment was studied in order to gain a better understanding of the epidemiology of this pathogen, notably isolated from two distinct shrimp disease complexes. Molecular typing using two different techniques, arbitrarily primed PCR (AP-PCR) and multi-locus sequence typing (MLST), studied together with experimental pathology data allowed a relevant epidemiological insight into this possibly emerging pathogen. Additionally, results obtained with the two molecular typing techniques were congruent and allowed discriminating the strains associated with the "Summer Syndrome" from strains isolated from other contexts, especially the other shrimp...

In order to improve oyster culture, many research programs try and propose new methods aiming at improving genetically the product performances and quality (selection, polyploidisation). As for terrestrial animal production, the continuity of the different works as well as the spreading of the genetical progress would be made easier by the development of conservation techniques of the selected genotypes. Cryopreservation, which is often used in selection programs of terrestrial animals or fish farming species, is not always available for bivalves. This technique would enable the conservation of the gametes of individuals selected for their high growth potential, their resistance to diseases and/or their quality for a smaller cost and less risks than the...

Development of an antiseptic procedure to improve retention rates during the grafting process of black-lip oyster, Pinctada margaritifera ArchiMer
Cochennec, Nathalie; Haffner, Philippe; Levy, Peva; Saulnier, Denis; Fougerouse, A.; Langy, Sandra.
In French Polynesia, black pearl cultivation now takes place on about 30 of the 118 atolls or islands. This activity has largely replaced copra, coconut oil and fishing as a source of income. Around 1,000 pearl farms, generally small ones of less than 5 ha, are registered. The pearl farms are mainly owned by families. In 1998, about 7,000 persons were employed in theentire pearl industry, from production to retailing. In value, this activity represents the first exportation of French Polynesian products. From 1988 to date, exports increased exponentially before leveling. Starting with less than two kilograms in 1972, exports have now stabilized at Il tons of "black" pearls.

Modulations of the interactions between pacific cupped oyster Crassostrea gigas and Vibrio according to bacterial virulence and to genetic and physiological status of the host : Methodology and first results ArchiMer
De Decker, Sophie; Normand, Julien; Duperthuy, Marylise; Boudry, Pierre; Saulnier, Denis.
Rearing of Crassostrea gigas is the most economically important aquaculture activity in France. Bacteria belonging to Vibrio genus constitute one of the most abundant bacterial group in marine ecosystems. Two Vibrio species, V. splendidus and V. aestuarianus, were associated with many cases of mortality events in reared C. gigas spat and juvenile oysters, commonly during summer. This summer mortality syndrome has been extensively documented as the result of complex interactions between pathogens, host and environmental conditions. This work aims to study the Vibrio-host interactions and their modulations according to the virulence of pathogens and the genetic and/or physiological parameters of the host. These factors will be characterized by in vitro and...

Pearl formation: Persistence of the graft during the entire process of biomineralization ArchiMer
Arnaud, Sophie; Goyard, Emmanuel; Vonau, Vincent; Herbaut, C; Prou, Jean; Saulnier, Denis.
Most bivalves species of the genus Pinctada are well known throughout the world for production of white or black pearls of high commercial value. For cultured pearl production, a mantle allograft from a donor is implanted into the gonad of a recipient oyster, together with a small inorganic bead. Because of the dedifferentiation of cells during the first steps of the host oyster's immunological reaction, so far the fate of the graft and its exact role in the process of pearl formation could not be determined via classical histological methods. Here we report the first molecular evidence of the resilience of the graft in the recipient organism by showing that cells containing genome from the donor are still present at the end of pearl formation. These...

Characterization of actin genes in Bonamia ostreae and their application to phylogeny of the Haplosporidia ArchiMer
Lopez Flores, L; Suarez Santiago, V; Longet, D; Saulnier, Denis; Chollet, Bruno; Arzul, Isabelle.
Bonamia ostreae is a protozoan parasite that infects the European flat oyster Ostrea edulis, causing systemic infections and resulting in massive mortalities in populations of this valuable bivalve species. In this work, we have characterized B. ostreae actin genes and used their sequences for a phylogenetic analysis. Design of different primer sets was necessary to amplify the central coding region of actin genes of B. ostreae. Characterization of the sequences and their amplification in different samples demonstrated the presence of 2 intragenomic actin genes in B. ostreae, without any intron. The phylogenetic analysis placed B. ostreae in a clade with Minchinia tapetis, Minchinia teredinis and Haplosporidium costale as its closest relatives, and...

Combination of a pesticide exposure and a bacterial challenge: In vivo effects on immune response of Pacific oyster, Crassostrea gigas (Thunberg) ArchiMer
Gagnaire, Beatrice; Gay, Melanie; Huvet, Arnaud; Daniel, Jean-yves; Saulnier, Denis; Renault, Tristan.
To assess the impact of pollution induced by pesticides on Pacific oyster, Crassostrea gigas, health in France, in vivo effects of combined pesticide exposure and bacterial challenge on cell activities and gene expression in hemocytes were tested using flow cytometry and real-time PCR. As a first step, an in vivo model of experimental contamination was developed. Pacific oysters were exposed to a mixture of eight pesticides (atrazine, glyphosate, alachlor, metolachlor, fosetyl-alumimium, terbuthylazine, diuron and carbaryl) at environmentally relevant concentrations over a 7-day period. Hemocyte parameters (cell mortality, enzyme activities and phagocytosis) were monitored using flow cytometry and gene expression was evaluated by real-time PCR (RT-PCR)....

Construction of a Vibrio splendidus Vsm metalloprotease mutant using a novel counter-selectable suicide vector ArchiMer
Leroux, Frederique; Binesse, Johan; Saulnier, Denis; Mazel, Didier.
splendidus is a dominant culturable Vibrio in seawater, but strains related to this group are also associated with mortalities in a variety of marine animals. The determinants encoding the pathogenic properties of these strains are still poorly known, however, the recent sequencing of the genome of V. splendidus LGP32, an oyster pathogen, offers the opportunity to decipher the basis of the virulence properties by the disruption of candidate genes. We have developed a novel suicide vector based on the pir-dependant R6K replicative origin, which can be transferred by RP4 based conjugation to, potentially, any Vibrio strain, and which also carries the plasmid F toxin ccdB gene under the control of the PBAD promoter. We demonstrate that this genetic system...
